What retatrutide is

Retatrutide was first coded LY3437943 by Eli Lilly. Its discovery paper calls it a triple agonist peptide, meaning one molecule switches on the glucagon receptor, the GIP receptor and the GLP-1 receptor.

In cell tests the team found balanced glucagon and GLP-1 receptor activity, with stronger action at the GIP receptor. That three-way design is what sets it apart from older single or dual receptor peptides.

What the published studies examined

Early work in obese mice looked at body weight, blood sugar control and energy use. A first trial in people then measured safety, tolerability and how long the peptide stays in the blood.

Two phase 2 trials followed in 2023. One enrolled adults with obesity and the other enrolled adults with type 2 diabetes, and both tracked side effects and safety across several study arms.

In 2026 the New England Journal of Medicine published TRIUMPH-1, a phase 3 trial in adults with obesity. It also looked at knee pain in people with knee osteoarthritis and at breathing pauses during sleep in a smaller group.

None of this makes retatrutide an approved medicine. It is still an investigational compound.

Storage and handling of the powder

Freeze-dried peptides last longest as a dry cake in a tightly closed vial kept below -15 °C. Colder storage is better for long holding times.

Peptide powder pulls water from the air, and that moisture lowers stability. Warm a cold vial to room temperature before you open it so condensation does not form inside, and keep it out of strong light.

Retatrutide's backbone carries a glutamine residue, one of the amino acids that suppliers flag as giving a shorter shelf life in solution, so peptide labs advise against long storage once dissolved.

Regulatory status

In Australia

Retatrutide is not approved for supply in Australia, and the TGA includes it in its list of unapproved peptide products. The TGA says it is unlawful to import, advertise, manufacture and supply unapproved peptide products in Australia. NSW Health gives the same warning.

On 19 June 2026 the Victorian Department of Health issued an alert titled 'Toxicity linked to unapproved peptide product labelled Retatrutide'.

Is retatrutide approved in Australia?

No. It is not on the Australian Register of Therapeutic Goods, and it appears on the TGA's unapproved peptide list. Glow Labs supplies it for laboratory research only.

Are there clinical trials of retatrutide?

Yes. Phase 2 trials were published in 2023 in adults with obesity and in adults with type 2 diabetes, and the phase 3 TRIUMPH-1 trial was published in 2026. ClinicalTrials.gov lists 6 retatrutide studies with Australian sites (checked 26 October 2026).

What is the difference between retatrutide and tirzepatide?

Tirzepatide acts on two receptors, GIP and GLP-1. Retatrutide adds a third, the glucagon receptor. Tirzepatide is an approved medicine in Australia; retatrutide is still investigational.
